Analysis of larval oyster grazing by flow cytometry ArchiMer

The ingestion of 8 algal species by oyster larvae (Crassostrea gigas Thunberg, 1793) was measured by flow cytometry (FCM). In a preliminary experiment, cell size (estimated by light scatter) and chlorophyll fluorescence of 30 algal species were evaluated to select those species which could be mixed together and still be easily discriminated by FCM. Grazing experiments were carried out over 48 h with 6 and 15-day old larvae fed on 3 algal mixtures, each containing 3 different algal species. The concentration of each species was estimated at 0, 6, 12, 24 and 48 h by FCM. Grazing pressure on a given algal species was dependent upon the age of the larvae, the time of the day and the composition of the mixture. Grazing rates of older larvae were about twice...

Environmental and species features which lead to increases in cell density of the D.S.P. Dinoflagellate Dinophysis in inshore and offshore waters of the île de Ré - île d'Oléron area, France (cruises "Dinopertuis89"). Four weekly sampling series have been carried out, within 10 May – 31 May 1989, at 24 stations located from inshore to offshore waters of the "île de Ré" - "île d’Oléron" area, with the aim to investigate (i) whether Dinophysis increases in cell density occur first in offshore waters and then, eventually, are pushed to the coast, or develop simultaneously in the whole area, and (ii) whether there are some environmental conditions which are related to Dinophysis blooms. Dinophysis spp. cells concentrated from nature for experimental purposes, using size fractionation and reverse migration ArchiMer

A method based on size fractionation plus reverse phototactic migration was used to collect large Dinophysis spp.-dominated populations. Cells were active and survived handling and transportation. Experiments involving various treatments for establishing permanent cultures are described; none have yet been successful.
